What are the main modes and application areas of super preparative chromatography?

Preparative chromatography is a chromatographic system with the purpose of separating pure substances. Analytical chromatography is a chromatography system with the purpose of analyzing sample component information. Based on their different purposes, preparative chromatography has many different characteristics than analytical chromatography. There are three commonly used chromatography modes in preparative chromatography: low pressure, medium pressure, and high pressure to cope with the separation of different types and different purity requirements of samples while reducing costs as much as possible.


Low pressure: Generally speaking, low pressure is suitable for easy separation of compounds, but long separation time can easily cause the decomposition of sensitive compounds.

Medium pressure: Medium pressure generally uses a constant flow pump to provide a constant mobile phase flow rate. The filler particles are smaller, the resolution is higher, and it can withstand higher pressure and faster flow rate.

High pressure: High pressure is usually used in the later stages of separation preparation and is used for samples that are difficult to separate. The purity can even reach 99.9%.


Compared with traditional purification methods (such as distillation and extraction), preparative chromatography is a more effective separation method and is therefore widely used in the extraction and purification of samples and products. As the demand for high-purity components continues to increase in the fields of synthesis, phytochemistry, biochemistry and pharmaceuticals, the application fields of preparative liquid chromatography are also rapidly expanding. Currently, laboratory-grade preparative liquid chromatography is mainly used in the following areas:

1. Natural medicinal chemistry, traditional Chinese medicine chemistry;

2. Protein purification;

3. Purification of drug impurities;

4. Preparation of chiral compounds;

5. Industrial hemp extraction;


6. High purity fish oil extraction, etc.


The advantages of super preparative chromatography mainly include the following:

1. Large sample loading capacity and high filler utilization rate;

2. Some reagents are internally circulated, and the reagent usage is low, less than 1/2 of the reagent usage in traditional processes;

3. The product yield is high, basically around 95%;

4. The purity can be above 99%, and the impurities can be controlled below 0.2%;

5. There is a 1000mm diameter super preparative chromatograph that can verify relevant process parameters on site.
